your Neopixel should receive 5V from a dedicated PSU (as you don't state the number of LEDs in use) as they will consume quite some current.
100nF is a decoupling capacitor for the 3.3V supply of the GPIO expander.
I have a 10 Ampère 5V power supply in place and I actually planned to power the Pixels together with the Pi from it. I will run about 130 LEDs. Do I understand that correct, that I should use two different power supplies, one with 5V for the LEDs and another one with 3.3V for the Pi and Electronics?
So far you did not mention any details on your PSU.
you should be careful with your 5V@10A PSU as you still seem to not have understood that the Pi does not run from 3.3V; power supply to the Pi is 5V only --> you feed 5V into the Pi and 5V into the Neopixel string.
